The 45-ranked Americane prevailed in an hour and a half, at the expense of the 22-year-old Ontarian.
Champion at Flushing Meadows in 2019, Andreescu is 54e at the WTA.
Rogers earned all three breaks in the game, in part because her rival committed all five double faults in the game.
Rogers will face favorite Maria Sakkari, the third racquet on the planet.
Canadian Gabriela Dabrowski will be in action on Wednesday in doubles.
She and Giuliana Olmos of Mexico will face Emina Bektas and Kaitlyn Christian of the United States.
Dabrowski and Olmos are the second seeds of the tournament.
