Playing four groups ahead of Cantlay, Spieth birdied at 18and hole on the Harbor Town course and he shot 66 (-5).
Cantlay pared the last pennant to put up a 68 and tie Spieth with a minus-13 overall.
When Spieth completed his round, he was one stroke behind Shane Lowry, but the latter committed a double bogey on 14and hole. Lowry posted a 69 card and finished the tournament with a cumulative score of minus-12.
In overtime, Cantlay sent his ball into the sand trap. He placed his ball 35 feet from the cup and missed his par attempt.
Three strokes behind Harold Varner III before the start of his final round, Spieth birdied on both par-5s of the front nine.
Spieth recorded a 13and victory on the PGA Tour and a second consecutive victory on Easter Sunday. He ended a four-year winless drought by winning the Valero Texas Open in 2021.
Next year, the final round of the Masters Tournament will take place on Easter Sunday.
Cam Davis (63), Varner (70), JT Poston (64), Cameron Young (66), Sepp Straka (68) and Matt Kuchar (68) joined Lowry tied for third.
By virtue of a round of 69, Corey Conners was the tournament’s highest-ranked Canadian, tied for 12and square. His compatriots Adam Hadwin (69) and Adam Svensson (71) both took 26and rank while Roger Sloan (73) settled for a tie in 59and square.
